چکیده

Transcriptional gene silencing is mediated by various epigenetic modifications including DNA methylation, histone modifications and recruitment of binding proteins that reads the methyl and other modification marks. The order in which these modifications occur followed by repressor protein recruitment remains contentious. Here, using purified protein components, we show that mammalian RNA polymerase II (RNA Pol II) is involved in DNA methylation control. DNA (cytosine-5) methyltransferase 1 (DNMT1) colocalizes, directly interacts and binds to the phosphorylated Cterminal repeat domain (CTD) of Rpb1, a major structural subunit of RNA Pol II. The association of RNA Pol II with DNMT1 during transcription enhances DNA methylation, and methylated DNA doesn’t affect in vitro transcription. Addition of methyl CpG binding protein 2 (MeCP2), inhibited in vitro transcription of DNA in a methylation dependent manner, suggesting a possible mechanism for RNA Pol II coupled transcriptional silencing mediated by DNMT1 and

Lsh Mediated RNA Polymerase II Stalling at HoxC6 and HoxC8 Involves DNA Methylation

DNA cytosine methylation is an important epigenetic mechanism that is involved in transcriptional silencing of developmental genes. Several molecular pathways have been described that interfere with Pol II initiation, but at individual genes the molecular mechanism of repression remains uncertain. Here, we study the molecular mechanism of transcriptional regulation at Hox genes in dependence of...

O-11: N-a-acetyltransferase 10 Protein Regulates DNA Methylation and Embryonic Development

Background Genomic imprinting is a heritable and developmentally essential phenomenon by which gene expression occurs in an allele-specific manner1. While the imprinted alleles are primarily silenced by DNA methylation, it remains largely unknown how methylation is targeted to imprinting control region (ICR), also called differentially methylated region (DMR), and maintained. Here we show that ...

Mechanisms of HDA6-mediated rRNA gene silencing: suppression of intergenic Pol II transcription and differential effects on maintenance versus siRNA-directed cytosine methylation.

The Arabidopsis histone deacetylase HDA6 is required to silence transgenes, transposons, and ribosomal RNA (rRNA) genes subjected to nucleolar dominance in genetic hybrids. In nonhybrid Arabidopsis thaliana, we show that a class of 45S rRNA gene variants that is normally inactivated during development fails to be silenced in hda6 mutants. In these mutants, symmetric cytosine methylation at CG a...

Detection of Pol IV/RDR2-dependent transcripts at the genomic scale in Arabidopsis reveals features and regulation of siRNA biogenesis.

Twenty-four-nucleotide small interfering (si)RNAs are central players in RNA-directed DNA methylation (RdDM), a process that establishes and maintains DNA methylation at transposable elements to ensure genome stability in plants. The plant-specific RNA polymerase IV (Pol IV) is required for siRNA biogenesis and is believed to transcribe RdDM loci to produce primary transcripts that are converte...

RNA-directed DNA methylation and plant development require an IWR1-type transcription factor.

RNA-directed DNA methylation (RdDM) in plants requires two RNA polymerase (Pol) II-related RNA polymerases, namely Pol IV and Pol V. A genetic screen designed to reveal factors that are important for RdDM in a developmental context in Arabidopsis identified DEFECTIVE IN MERISTEM SILENCING 4 (DMS4). Unlike other mutants defective in RdDM, dms4 mutants have a pleiotropic developmental phenotype. ...
